1562-ci ildə dünyaya gəldi. Atası Mantuya hersoqu Quqlielmo Qonzaqa, anası isə Müqəddəs Roma imperatoru I Ferdinandın qızı Elyanoradır. Atasının vəfatından sonra 1587-ci ildə taxta çıxdı.
1581-ci ildə Marqarita Farneze ilə evlənsə də, 2 il sonra boşandılar və övladları olmadı. İkinci evliliyi isə 29 aprel 1584-cü ildə xalası qızı Elyanora Mediçi ilə baş tutdu. Cütlüyün bu nigahdan 6 övladı dünyaya gəlsə də, bunlardan biri uşaq yaşlarında vəfat etdi.
